an analog of GM-CSF with a mutated residue 21 (glutamic acid exchanged for arginine) in the first helix of GM-CSF, which is known to play a critical role in both biological activity and high-affinity receptor binding (see also: Muteins). E21R binds to the GM-CSF receptor-alpha chain with an affinity similar to that of wild-type GM-CSF and has lost high-affinity binding to the GM-CSF receptor complex consisting of the alpha chain and the common beta chain.
E21R antagonizes the proliferative effect of GM-CSF on the erythroleukemic cell line TF-1 and primary acute myeloid leukemias, and also superoxide production by neutrophils stimulated with GM-CSF
