This antigen is identified by a monoclonal antibody of the same name. The gene encoding the B-B4 antigen has been cloned and encodes CD138 (syndecan-1) (Wijdenes et al, 1996).
The antibody specifically identifies human plasma cells. It strongly reacts with multiple myeloma cell lines and malignant plasma cells of all tumor samples of the multiple myeloma patients tested. B-B4 does not react with any peripheral blood, bone marrow or tonsil cells (Wijdenes et al, 1996).
B-B4 recognizes both the intact molecule and the core protein of Syndecan-1. It shows a comparable reactivity in flow cytometry and immunoblotting with antibodies 1D4 and Mi15
